چکیده

The energetics of ATP’ hydrolysis to ADP and P have been studied previously by combining either thermal (1, 2) or free energy (3-6) data for a number of chemical reactions according to the usual procedures of chemical thermodynamics. Such calculations, however, suffer from the fact that the uncertainties in each measurement are compounded in the final result. The ATPase properties of myosin offer a more direct method for measurement of the enthalpy change on hydrolysis of ATP. In the present paper we submit measurements which, together with data on the heat of neutralization of the buffer system employed, show that the heat evolved during hydrolysis of ATP is approximately half the value 12 kilocalories per mole which has been hitherto accepted.

The Effect of Vitrification and in vitro Culture on the Adenosine Triphosphate Content and Mitochondrial Distribution of Mouse Pre-Implantation Embryos

Background: The mitochondria are an important source of adenosine triphosphate (ATP) production in pre-implantation embryo. Therefore, the objective of this study was to investigate the effect of vitrification and in vitro culture of mouse embryos on their mitochondrial distribution and ATP content. Methods: The embryos at 2-PN, 4-cell and blastocyst stages were collected from the oviduct of st...

Comparative effects of four uncouplers on the enthalpy change for mitochondrial oxidation of succinate by ferricyanide.

The enthalpy changes for phosphorylating and nonphosphorylating mitochondrial oxidation of succinate by ferricyanide were studied by heatburst microcalorimetry. The enthalpy change observed with a nonphosphorylating system involving sonically disrupted mitochondria agreed with the calculated enthalpy change. The enthalpy changes for phosphorylating and uncoupled systems involving intact mitocho...

A transient kinetic study of enthalpy changes during the reaction of myosin subfragment 1 with ATP.

1. The enthalpy changes during individual reaction steps of the myosin subfragment 1 ATPase were studied with the use of a new stopped-flow calorimeter [Howarth, Millar & Gutfreund (1987) Biochem. J. 248, 677-682]. 2. At 5 degrees C and pH 7.0, the endothermic on-enzyme ATP-cleavage step was observed directly (delta H = +64 kJ.mol-1). 3. ADP binding is accompanied by a biphasic enthalpy change....

An Adenosine Triphosphate Bioluminescence Method for Evaluating the Microbial Contamination of the Salad-Preparing Tables and Salad-Serving Dishes in Restaurants of Mashhad City, Iran

Introduction: Consumption of vegetable products is increasing commonly in the world because they are recognized as an important source of nutrients, vitamins, and fiber for humans. Salads are among the most widely used foods that are also known as the most contaminated foods in restaurants. This study was conducted to determine the microbial contamination of salad-preparing tables and salad-ser...

Comparing Human Sperm Quality Preserved at Two Different Temperatures; Effect of Trolox, Coenzyme Q10 and Extracellular Adenosine Triphosphate

Cooling method was proposed to maintain the sperm quality for several days. Nevertheless, during this procedure, sperm is encountered to “cold shock”, and its quality decreases time-dependently. This study was designed to improve the in vitro sperm preservation methods. Thirty normal semen samples were examined in Shiraz, Iran, 2017. Fifteen samples were incubated at 22-27 °C and 15 samples wer...
